Active caspase-3 expression levels as bioindicator of individual radiosensitivity Anais da ABC (AABC)
SANTOS,NEYLIANE F.G. DOS; SILVA,RAFAEL F.; PINTO,MARCELA M.P.L.; SILVA,EDVANE B. DA; TASAT,DEBORAH R.; AMARAL,ADEMIR.
ABSTRACT Several molecules and events involved in cell response to radiation-induced damage have been investigated towards a personalized radiotherapy. Considering the importance of active caspase-3 in the proteolytic cascade that ensures radiation-induced apoptosis execution, this research was designed to evaluate the expression levels of this protein as a bioindicator of individual radiosensitivity. Peripheral blood samples of 10 healthy individuals were gamma-irradiated (cobalt-60 source) with 1, 2 and 4 Gy (control: non-irradiated samples), and active caspase-3 expression levels were measured in lymphocytes, by flow cytometry, ex vivo and after different times of in vitro incubation (24, 48 and 72 hours). Short-term incubation of 24 h was the most...

Caspase-3 activation and increased procollagen type I in irradiated hearts Anais da ABC (AABC)
Ferreira-Machado,Samara C.; Salata,Camila; Rocha,Nazareth N.; Corrêa,Alexandre F.S.; Côrte-Real,Suzana; Peregrino,Antônio A.F.; Campos,Vera M.A. de; Andrade,Cherley B.V.; Bernardo-Filho,Mario; Cabral-Neto,Januário B.; Dealmeida,Carlos E..
The caspase-3-cleaved presence was evaluated in this study in the heart of irradiated rats, during the decline of ventricular function. Female Wistar rats were irradiated with a single dose of radiation (15 Gy) delivered directly to the heart and the molecular, histological and physiological evaluations were performed at thirteen months post-irradiation. The expressions of procollagen type I, TGF-ß1 and caspase-3-cleaved were analyzed using Western blotting. Cardiac structural and functional alterations were investigated by echocardiography and electron microscopy. In the irradiated group, the levels of procollagen type I, TGF-ß1 and caspase-3-cleaved are increased. Significant histological changes (degeneration of heart tissue and collagen deposition) and...

Chicken Meat Submitted to Gamma Radiation and Packed with or without Oxygen Rev. Bras. Ciênc. Avic.
Pelicia,K; Garcia,EA; Molino,AB; Santos,GC; Vieira Filho,JA; Santos,TA; Berto,DA.
The objective of this study was to evaluate the effects on gamma radiation levels on the physical and microbiological characteristics of chicken breast meat. A completely randomized experimental design in a 4x2x3 factorial arrangement was adopted. Treatments consisted of four radiation concentrations (0, 2, 4, or 8kGy), two package sealing methods (with or without vacuum), and three storage times (01, 07, or 14 days), with ten replicates each, totaling 240 chicken breast fillets. Packaging and radiation had no influence (p>0.05) on chicken breast meat pH, water retention capacity, or presence of Salmonella spp. Breast fillets not submitted to radiation and vacuum packed presented higher water retention capacity (p<0.05) than those radiated at 4kGy...

Cytogenetic and cytomolecular delayed damage induced in human fibroblasts by low X-ray doses JBAG
Ponzinibbio,MV; Crudeli,C; Peral García,P; Seoane,A.
During the last decade several reports have provided evidence on the non-targeted effects of ionizing radiation. In this study, we have analyzed the delayed DNA damage induced by ionizing radiation in MRC-5 Human fibroblasts cultured in vitro. MRC-5 cells were irradiated with 10 and 50 milligrays (mGy) and analyzed immediately after exposure, and at passages 6 and 12 post-radiation by means of comet assay, micronucleus analysis and γ-H2AX foci formation. Cytogenetic and cytomolecular damage was analysed at every sample point comparing irradiated and control cells. No significant damage differences were found among samples from all passages of treatments. Our results suggest that even at low doses, ionizing radiation could induce delayed damage....

DETOXIFICATION OF SNAKE VENOM USING IONIZING RADIATION J. Venom. Anim. Toxins
ROGERO,J.R.; NASCIMENTO,N..
It is generally recognized that energy absorbed from ionizing radiation (gamma rays) can inactivate biological material in two ways. A direct effect occurs when the primary event, i. e., ionization, is produced in the molecule itself. This is the case when a compound is irradiated in dry state. When a compound is irradiated in a solution, the indirect effect joins the direct(9). Since water is the most abundant constituent of biological material, it is important to consider the species produced by excitation and ionization of water itself, and the reaction of these species with the target molecules of biological importance. This indirect effect results from the reactions among the studied molecules and the products of radiation interaction with water or...

Effect of ionizing radiation onVibrio bacteria in Crassostrea virginica (American oyster) ArchiMer
Eberiel, D; Dooley, K; Langton, L; Malleth, J.
The presence of Vibrio bacteria in raw shellfish poses a significant health hazard to the consuming public. These human pathogens are natural, seasonal components of the microbial flora of shellfish which can result in primary septicemic or diarrheal fatal infections. Existing shellfish depuration neither targets nor eliminates the Vibrio bacteria. For this reason, additional purification processes are necessary. In this study, the effect of low dose gamma radiation on the inactivation ofVibrio cholerae (01 and non 01) and vulnificus is investigated. Log decrement (D10) values ranging only to a high of 0.09 KGy in the Vibrio strains support the hypothesis that food irradiation technology can provide an additional effective and efficient shellfish...

Evaluation of radioinduced damage and repair capacity in blood lymphocytes of breast cancer patients BJMBR
Nascimento,P.A.; da Silva,M.A.; Oliveira,E.M.; Suzuki,M.F.; Okazaki,K..
Genetic damage caused by ionizing radiation and repair capacity of blood lymphocytes from 3 breast cancer patients and 3 healthy donors were investigated using the comet assay. The comets were analyzed by two parameters: comet tail length and visual classification. Blood samples from the donors were irradiated in vitro with a 60Co source at a dose rate of 0.722 Gy/min, with a dose range of 0.2 to 4.0 Gy and analyzed immediately after the procedure and 3 and 24 h later. The basal level of damage and the radioinduced damage were higher in lymphocytes from breast cancer patients than in lymphocytes from healthy donors. The radioinduced damage showed that the two groups had a similar response when analyzed immediately after the irradiations. Therefore, while...

Hepatocytes, rather than leukocytes reverse DNA damage in vivo induced by whole body y-irradiation of mice, as shown by the alkaline comet assay Biol. Res.
PINCHEIRA,JUANA; CARRERA,PILAR; MARCELAIN,KATHERINE; DE LA TORRE,CONSUELO.
DNA damage repair was assessed in quiescent (G0) leukocytes and in hepatocytes of mice, after 1 and 2 hours recovery from a single whole body y-irradiation with 0.5, 1 or 2 Gy. Evaluation of single-strand breaks (SSB) and alkali-labile sites together were carried out by a single-cell electrophoresis at pH>13.0 (alkaline comet assay). In non-irradiated (control) mice, the constitutive, endogenous DNA damage (basal) was around 1.5 times higher in leukocytes than in hepatocytes. Irradiation immediately increased SSB frequency in both cell types, in a dose-dependent manner. Two sequential phases took place during the in vivo repair of the radio-induced DNA lesions. The earliest one, present in both hepatocytes and leukocytes, further increased the SSB...

In vitro apprach to the study of chronic exposure to low does of X-rays JBAG
Ponzinibbio,M.V; Peral-García,P; Seoane,A.
The present research was undertaken in order to study the effect of repeated low doses of radiation on two different cell lines from the same species. An in vitro model test was developed to avoid the influence of the confounding factors affecting epidemiological studies and to simulate a chronic exposure (50 mSv of x-rays during ten consecutive days). Comet assay and early apoptosis were analyzed immediately after exposure and after chronic irradiation. Sequential irradiation induced an increase of cells showing DNA damage. Index Damage was higher than that of the controls in both cell lines immediately after exposure and after chronic irradiation. These differences between exposed and control cells were statistically significant only for the transformed...

Influence of Ionizing Radiation on Biodegradable Foam Trays for Food Packaging Obtained from Irradiated Cassava Starch BABT
Brant,Antônio Jedson Caldeira; Naime,Natália; Lugão,Ademar Benévolo; Ponce,Patrícia.
ABSTRACT Biodegradable polymers, such as starch, cellulose, and derivatives thereof, are being studied to produce innovative packaging in the most diverse shapes (films, bags, trays, bottles, etc.) to attend this current market trend. The aim of this work was to produce foam trays from cassava starch for food packaging via extrusion followed by thermoforming process. Their formulations were based on unirradiated and γ-irradiated starches at diverse radiation absorbed doses (in kGy) in order to evaluate the influence of the irradiated starches on the physical properties of the trays. Water absorption results showed an irregular increase with the increase of the absorbed dose ( for example, 26.32% and 39.84% for the trays based on unirradiated starch 0 kGy...

Ionizing radiation-induced DNA injury and damage detection in patients with breast cancer Genet. Mol. Biol.
Borrego-Soto,Gissela; Ortiz-López,Rocío; Rojas-Martínez,Augusto.
Abstract Breast cancer is the most common malignancy in women. Radiotherapy is frequently used in patients with breast cancer, but some patients may be more susceptible to ionizing radiation, and increased exposure to radiation sources may be associated to radiation adverse events. This susceptibility may be related to deficiencies in DNA repair mechanisms that are activated after cell-radiation, which causes DNA damage, particularly DNA double strand breaks. Some of these genetic susceptibilities in DNA-repair mechanisms are implicated in the etiology of hereditary breast/ovarian cancer (pathologic mutations in the BRCA 1 and 2 genes), but other less penetrant variants in genes involved in sporadic breast cancer have been described. These same genetic...

L'ionisation des produits de la pêche ArchiMer
Kodo, Jean-louis.
Fishes and fishery products are seafoods which have a nutritive value with a high proteins rate. Fishery products ionization has four steps: the radappertization (sterilization of the seafood), the radicidation (destruction of the pathogen germs), the radurization (food conservation and microbial flora reduction), the desinsectization (insects and parasites elimination). Fishery products quality control is very important

Long-term correlation of the electrocorticogram as a bioindicator of brain exposure to ionizing radiation BJMBR
Aguiar,L.A.A.; Silva,I.M.S.; Fernandes,T.S.; Nogueira,R.A..
Understanding the effects of radiation and its possible influence on the nervous system are of great clinical interest. However, there have been few electrophysiological studies on brain activity after exposure to ionizing radiation (IR). A new methodological approach regarding the assessment of the possible effects of IR on brain activity is the use of linear and nonlinear mathematical methods in the analysis of complex time series, such as brain oscillations measured using the electrocorticogram (ECoG). The objective of this study was to use linear and nonlinear mathematical methods as biomarkers of gamma radiation regarding cortical electrical activity. Adult Wistar rats were divided into 3 groups: 1 control and 2 irradiated groups, evaluated at 24 h...

Low doses of gamma ionizing radiation increase hprt mutant frequencies of TK6 cells without triggering the mutator phenotype pathway Genet. Mol. Biol.
Ayres,Flávio Monteiro; Cruz,Aparecido Divino da; Steele,Patricia; Glickman,Barry W..
The TK6 lymphoblastoid cell line is known to be mismatch repair (MMR) and p53 proficient. Deficiency in MMR results in a mutator phenotype characterized by microsatellite instability (MSI) and increased hprt mutant frequency (MF). Increased hprt MF is also a biomarker of effect for exposure to ionizing radiation. In order to test if a mutator phenotype could be induced by low doses of gamma ionizing radiation, an hprt cloning assay and a MSI investigation were performed after radiation exposure. The spontaneous MF was 1.6 x 10-6. The groups exposed to 0.2, 0.5 and 1.0 Gy had hprt MFs of 2.3, 3.3 and 2.2 x 10-6, respectively. The spontaneous MSI frequency per allele in non-selected cells was 5.4 x 10-3, as evidenced at the loci D11S35, nm23-H1, D8S135 and...

Low doses of gamma radiation in the management of postharvest Lasiodiplodia theobromae in mangos BJM
Santos,Alice Maria Gonçalves; Lins,Severina Rodrigues Oliveira; Silva,Josenilda Maria da; Oliveira,Sônia Maria Alves de.
The postharvest life of mango is limited by the development of pathogens, especially fungi that cause rot, among which stands out the Lasiodiplodia theobromae. Several control methods have been employed to minimize the damages caused by this fungus, chemical control can leave residues to man and nature; physical control by the use of gamma radiation in combination with modified atmosphere and cold storage. The use of gamma radiation helps to reduce the severity of the pathogen assist in the ripening process of fruits, even at low doses (0.25, 0.35 and 0.45 kGy) chemical properties such as pH, soluble solids, acid ascorbic, titratable acidity and also the quality parameters of the pulp showed no damage that are ideal for trade and consumption of mangoes....

Overexpression of rice F-box protein OsFBX322 confers increased sensitivity to gamma irradiation in Arabidopsis Genet. Mol. Biol.
Hwang,Jung Eun; Hwang,Sun-Goo; Jung,In Jung; Han,Sung Min; Ahn,Joon-Woo; Kim,Jin-Baek.
Abstract Ionizing radiation has a substantial effect on physiological and biochemical processes in plants via induction of transcriptional changes and diverse genetic alterations. Previous microarray analysis showed that rice OsFBX322, which encodes a rice F-box protein, was downregulated in response to three types of ionizing radiation: gamma irradiation, ion beams, and cosmic rays. In order to characterize the radiation-responsive genes in rice, OsFBX322 was selected for further analysis. OsFBX322 expression patterns in response to radiation were confirmed using quantitative RT-PCR. Transient expression of a GFP-OsFBX322 fusion protein in tobacco leaf epidermis indicated that OsFBX322 is localized to the nucleus. To determine the effect of OsFBX322...

Potential Radioprotective Effect of AT1 Receptor Antagonists Against Morphological and Ultrastructural Changes in the Testes Induced by Ionizing Radiation International Journal of Morphology
Schwarz,Franciele Cristina; Mansano,Naira da Silva; Chies,Agnaldo Bruno; Viani,Gustavo Arruda; Spadella,Maria Angélica.
Radiotherapy is a source of human exposure to ionizing radiation. This pure energy causes deleterious effects on tissues, which result from oxidative stress, a phenomenon in which there is the participation of the Renin-Angiotensin System (RAS). The male genital organs are extremely radiosensitive and the action of radiation in the testes can significantly affect spermatogenesis. In search of potential radioprotective for male genital system, this study investigated whether the AT1 receptor antagonists minimize radiation-induced damage to reproductive tissues, by decreasing oxidative stress. Male Wistar rats were divided into six groups: 0 Gray (Gy) (control), 5 Gy (single dose in the scrotal area), telmisartan, losartan, 5Gy+telmisartan and 5Gy+losartan....

Study of irradiated bothropstoxin-1 with60Co gamma rays: immune system behavior J. Venom. Anim. Toxins incl. Trop. Dis.
Caproni,P; Baptista,JA; Almeida,TL de; Passos,LAC; Nascimento,N.
Ionizing radiation has been successfully employed to modify the immunological properties of biomolecules. Very promising results were obtained when crude animal venoms, as well as isolated toxins, were treated with 60Co gamma rays, yielding toxoids with good immunogenicity. The achievement of modified antigens with lower toxicity and preserved or improved immunogenicity can be very useful. Ionizing radiation has already been proven to be a powerful tool to attenuate snake venom toxicity without affecting, and even increasing, their immunogenic properties. However, little is known about the modifications that irradiated molecules undergo and even less about the immunological response that such antigens elicit. In the present work, we investigated the...
